Explosives were being unloaded from a vehicle within the tunnel in Hunan, said the official Chinese news agency Xinhua.
The 2.4km tunnel is part of the Yanling-Rucheng expressway. Work is now under way to investigate the cause of the explosion.
Four other workers survived the blast.
